Associated gene: 200 corresponding target mRNAs, 9 candidate mRNA binding targets, HuR
Associated microRNA: hsa-miR-556-5p, hsa-miR-521, hsa-miR-1204, hsa-miR-146b-3p, hsa-miR-512-5p, hsa-miR-663b, hsa-miR-1258, hsa-miR-1233, hsa-miR-1296
Biological function: hsa_circ_0000520 may serve as a novel biomarker for gastric cancer, is associated with TNM stage and CEA expression, and may play an inhibiting role in gastric cancer occurrence and development.
Molecular mechanism: Predicted circRNA-miRNA-mRNA interaction network; the authors proposed that hsa_circ_0000520 may act through miRNA-mediated effects on gene expression in gastric cancer.
Biological pathway or process:

ceRNA regulation (other); other pathway/process (inhibits)

Detected method:
Q
M
Validation methods:

Microarray; RT-qPCR; Clinical Sample Validation; ROC Analysis; Bioinformatics Analysis

Clinical significance:

hsa_circ_0000520 might serve as a novel biomarker for gastric cancer; its tissue level was negatively associated with TNM stage, its plasma level was associated with CEA expression, and ROC analysis suggested diagnostic value especially in plasma.

Description:

This study identified hsa_circ_0000520 as significantly down-regulated in gastric cancer tissues, plasma, and gastric cancer cell lines. Lower hsa_circ_0000520 expression was associated with more advanced TNM stage and CEA positivity, suggesting potential diagnostic biomarker value and a possible inhibitory role in gastric cancer development. The proposed mechanism is a predicted circRNA-miRNA-mRNA regulatory network, but the molecular mechanism remains unvalidated experimentally.
