circRNA-disease information
Expression pattern:
UP
Associated gene: RAB27B, VEGFA
Associated microRNA: miR-607
Biological function: promotes angiogenesis; enhances migration, viability, invasiveness and tube formation of endothelial cells; promotes peritoneal dissemination
Molecular mechanism: Exosomal circPUM1 acts as a ceRNA to sponge miR-607, relieving repression of VEGFA and RAB27B; RAB27B enhances exosome release and VEGFA activates VEGF signaling in endothelial cells to promote angiogenesis.
Biological pathway or process:

angiogenesis (promotes); migration (promotes); invasion (promotes); proliferation (promotes); metastasis (promotes); ceRNA regulation (promotes); VEGF/VEGFR (promotes)

Detected method:
Q
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RT-qPCR; Clinical Sample Validation; Luciferase Reporter Assay; RNA Pull-Down; Transfection; CCK8; EdU Staining; Wound Healing Assay; Transwell Assay; Tube Formation Assay; Annexin V/PI Flow Cytometry; Western Blot; IF (Immunofluorescence); IHC (Immunohistochemistry); H&E Staining; In Vivo Animal Model; Bioinformatics Analysis

Clinical significance:

circPUM1 was positively correlated with advanced tumor stage and microvessel density (MVD) in ovarian cancer tissues.

Description:

circPUM1 is up-regulated in ovarian cancer and is associated with higher microvessel density. It is packaged into exosomes and transferred to endothelial cells to promote angiogenesis by sponging miR-607 and upregulating VEGFA and RAB27B, enhancing VEGF signaling and exosome release. Silencing circPUM1 with siRNA delivered by pH-responsive MXene nanosheets inhibits angiogenesis and peritoneal dissemination in vitro and in vivo.

Confidence score:

0.8007
