circ-FoxO3 is downregulated in the brains of APOE4 transgenic mice with AD-like neuropathology. Its overexpression reduces p-tau and Abeta accumulation, whereas knockdown worsens these aggregates. Mechanistically, circ-FoxO3 promotes FoxO3-mediated autophagy, suggesting therapeutic potential for APOE4-related neurodegenerative pathology.
