circRNA-disease information
Expression pattern:
DN
Associated gene: FoxO3, HuR, ubiquitin ligases, ribosomal subunits
Associated microRNA: miR-22, miR-543, miR-433
Biological function: Alleviates APOE4-induced brain damage, reduces p-tau and Abeta aggregation, promotes autophagy, and facilitates clearance of neurotoxic protein aggregates.
Molecular mechanism: circ-FoxO3 upregulates its parental gene FoxO3, activates autophagy, and enhances clearance of pathogenic p-tau and Abeta protein aggregates.
Biological pathway or process:

autophagy (promotes); other pathway/process (inhibits)

Detected method:
Q
Validation methods:

RT-qPCR; IF (Immunofluorescence); Transfection; In Vivo Animal Model; Western Blot

Clinical significance:

Potential therapeutic target for mitigating APOE4-related neuropathology.

Description:

circ-FoxO3 is downregulated in the brains of APOE4 transgenic mice with AD-like neuropathology. Its overexpression reduces p-tau and Abeta accumulation, whereas knockdown worsens these aggregates. Mechanistically, circ-FoxO3 promotes FoxO3-mediated autophagy, suggesting therapeutic potential for APOE4-related neurodegenerative pathology.

Confidence score:

0.414
