FGFR1, P65, FUS, AGO2, FGFR1 promoter |
| Associated microRNA: |
hsa-miR-4687-5p |
| Biological function: |
promotes PCa cell growth, migration, and invasion; promotes proliferation and DNA replication |
| Molecular mechanism: |
facilitates FGFR1 transcription by recruiting transcription activators P65/FUS and interacting with FGFR1 promoter; suppresses miR-4687-5p inhibitory effects on FGFR1 mRNA via competitive binding (miRNA sponge/ceRNA-like); co-exists with AGO2 in RISC |

| Clinical significance: |
Overexpression of circFGFR1int2 is significantly correlated with higher tumor grade, Gleason score, and PSA level, and is a significant unfavorable prognosticator for CRPC-free survival (CFS) (RR = 3.277, 95% confidence interval: 1.192-9.009; P = 0.021). |
| Description: |
circFGFR1int2 is up-regulated in prostate cancer and promotes tumor cell proliferation, migration, and invasion. It enhances FGFR1 expression by recruiting P65/FUS to the FGFR1 promoter to activate transcription and by sponging miR-4687-5p to relieve post-transcriptional repression of FGFR1. High circFGFR1int2 level correlates with higher grade/Gleason score/PSA and predicts worse CRPC-free survival. |
