circRNA-disease information
Expression pattern:
UP
Associated gene: hnRNPM, VEGFA, LGALS8, AKT1, NR4A1
Associated microRNA: -
Biological function: CircURI1 inhibits GC cell migration and invasion in vitro and represses GC metastasis in vivo; decreased circURI1 expression is associated with advanced tumor-node-metastasis stage and metastasis in GC patients.
Molecular mechanism: CircURI1 acts as a protein decoy that directly interacts with hnRNPM and sequesters hnRNPM to modulate alternative splicing of metastasis-related genes, including VEGFA.
Biological pathway or process:

migration (inhibits); invasion (inhibits); metastasis (inhibits); other pathway/process (other); VEGF/VEGFR (other); angiogenesis (other)

Detected method:

Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; RT-qPCR; RNA-seq; Northern Blot; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RIP (RNA Immunoprecipitation); CLIP; RNA Pull-Down; Transfection; Wound Healing Assay; Transwell Assay; Annexin V/PI Flow Cytometry; Cell Cycle Assay; In Vivo Animal Model; H&E Staining; Western Blot; Bioinformatics Analysis

Clinical significance:

CircURI1 has potential to act as a predictive biomarker or therapeutic target for GC; decreased circURI1 expression levels were significantly associated with advanced tumor-node-metastasis stage and metastasis in GC patients.

Description:

CircURI1 is a human URI1-derived circRNA that is up-regulated in gastric cancer tissues compared with adjacent nontumor tissues. Despite its increased expression, circURI1 functions as an anti-metastatic circRNA by inhibiting GC cell migration, invasion, and in vivo lung metastasis. Mechanistically, nuclear circURI1 binds and sequesters the RBP hnRNPM to modulate alternative splicing of metastasis-related genes, including VEGFA.
