circRNA-disease information
Expression pattern:
DN
Associated gene: FUS
Associated microRNA: hsa-miR-1265, hsa-miR-326
Biological function: Inhibits LPS-induced DNA damage and inflammatory responses in hDPSCs; alleviates pulpitis-associated DNA damage and inflammation in vivo.
Molecular mechanism: circ_0002456 binds FUS and restricts its nuclear export, suppressing NF-kappaB activation to mitigate DNA damage and inflammatory cytokine expression.
Biological pathway or process:

NF-kappaB (inhibits); inflammation (inhibits); apoptosis (not specified)

Detected method:
Q
S
Validation methods:

RNA-seq; RT-qPCR; Western Blot; RNase R Treatment; Sanger Sequencing; Back-Splice Junction PCR / divergent primers PCR; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; RNA Pull-Down; Transfection; IF (Immunofluorescence); In Vivo Animal Model; IHC (Immunohistochemistry)

Clinical significance:

-

Description:

circ_0002456 is down-regulated in pulpitis and in LPS-stimulated hDPSCs. It binds the RBP FUS, limits FUS nuclear export, and suppresses NF-kappaB activation, thereby reducing DNA damage (gamma-H2AX) and inflammatory cytokine expression; overexpression also attenuates these responses in a mouse pulpitis model.

Confidence score:

0.7929
