circRNA-disease information
Expression pattern:
UP
Associated gene: POLRMT, TIMM50
Associated microRNA: -
Biological function: promotes cardiac remodelling and heart failure phenotypes; decreases heart function; promotes fibrosis
Molecular mechanism: circRNA translation into SLC8a1-604; SLC8a1-604 translocates to mitochondria and binds POLRMT (with TIMM50), downregulating mitochondrial gene transcription and reducing ATP synthesis
Biological pathway or process:

mitochondrial function (inhibits); oxidative phosphorylation (inhibits); other pathway/process (other)

Detected method:
Q
Validation methods:

RT-qPCR; Western Blot; Nuclear-Cytoplasmic Fractionation; Transfection; RIP (RNA Immunoprecipitation); RNA Pull-Down; Co-IP; Luciferase Reporter Assay; Bioinformatics Analysis; RNA-seq; Clinical Sample Validation; In Vivo Animal Model; FISH / smFISH; ISH (In Situ Hybridization); IF (Immunofluorescence); ELISA

Clinical significance:

circSLC8a1 is significantly increased in heart failure patients and is suggested as a therapeutic target

Description:

In heart failure/cardiac pressure-overload remodelling, circSLC8a1 is up-regulated and exerts pathogenic effects mainly by being translated into SLC8a1-604. The translated protein interacts with TIMM50 and POLRMT in mitochondria to repress mitochondrial gene transcription and ATP production, contributing to impaired cardiac function and remodelling/fibrosis.

Confidence score:

0.7766
