circRNA-disease information
Expression pattern:
UP
Associated gene: PAX6
Associated microRNA: miR-132-3p
Biological function: Promotes RB cell proliferation, migration and invasion; inhibits apoptosis; promotes tumor growth in vivo; exosomal circRNF20 promotes malignant behaviors of RB cells.
Molecular mechanism: Acts as a miRNA sponge for miR-132-3p to regulate PAX6 (ceRNA mechanism); exosomal transfer increases circRNF20 level in recipient RB cells.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); EMT (promotes); metastasis (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Transfection; CCK8; EdU Staining; Colony Formation Assay; Annexin V/PI Flow Cytometry; Wound Healing Assay; Transwell Assay; Western Blot; Nuclear-Cytoplasmic Fractionation; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); RNA Pull-Down; In Vivo Animal Model; IHC (Immunohistochemistry); Survival Analysis; Clinical Sample Validation

Clinical significance:

circRNF20 elevation was linked to advanced TNM stage and poor overall survival rate; exosomal circRNF20 in serums might be a diagnostic marker for RB.

Description:

circRNF20 is up-regulated in retinoblastoma tissues/cells and is associated with advanced TNM stage and worse overall survival. Functionally, circRNF20 promotes RB cell proliferation, migration/invasion and EMT while inhibiting apoptosis, and it enhances tumor growth in a xenograft model. Mechanistically, circRNF20 sponges miR-132-3p to upregulate PAX6, and circRNF20 is also present in patient serum-derived exosomes that can enhance malignant phenotypes in RB cells.

Confidence score:

0.8616
