circPHF14 is upregulated in PDAC and promotes proliferation, migration/invasion, tumor growth and metastasis. It predominantly localizes to the cytoplasm and binds the RBP PABPC1 to stabilize WNT7A mRNA via the WNT7A 3’-UTR, activating Wnt/beta-catenin signaling and upregulating SNAI2 to drive EMT. EIF4E enhances circPHF14 biogenesis by binding specific flanking intronic regions in PHF14 pre-mRNA, and high circPHF14 predicts poor survival. |
