circRNA-disease information
Expression pattern:
UP
Associated gene: VCAM1, Ago2, RISC
Associated microRNA: miR-98-5p
Biological function: Circ-USP36 promotes ox-LDL-induced endothelial cell injury by reducing cell viability and promoting apoptosis and inflammatory responses in HUVEC cells.
Molecular mechanism: Circ-USP36 functions as a miRNA sponge for miR-98-5p, thereby increasing VCAM1 expression at the post-transcriptional level and promoting ox-LDL-induced HUVEC cell injury.
Biological pathway or process:

apoptosis (promotes); inflammation (promotes); proliferation (inhibits); ceRNA regulation (other)

Detected method:
Q
Validation methods:

RT-qPCR; Transfection; MTT; Annexin V/PI Flow Cytometry; ELISA; Western Blot; Bioinformatics Analysis; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation)

Clinical significance:

Circ-USP36/miR-98-5p/VCAM1 axis might be an underlying therapeutic target for endothelial dysfunction in AS; clinical sample validation was proposed as a future need, not performed in this study.

Description:

In an ox-LDL-induced HUVEC model of atherosclerosis-related endothelial injury, circ-USP36 is up-regulated and aggravates endothelial cell injury by inhibiting viability and promoting apoptosis and inflammation. Mechanistically, circ-USP36 sponges miR-98-5p to increase VCAM1 expression, forming a circ-USP36/miR-98-5p/VCAM1 regulatory axis that may represent a therapeutic target for endothelial dysfunction in AS.
