circRNA-disease information
Expression pattern:
UP
Associated gene: c-FOS
Associated microRNA: -
Biological function: Promotes glioma cell stemness, sphere-formation ability, ALDH activity, stemness marker expression, and tumor-initiating ability in vivo.
Molecular mechanism: c-FOS transcriptionally activates circRPPH1 expression by directly binding to the circRPPH1 promoter; circRPPH1 promotes glioma stemness dependent on c-FOS-mediated activation.
Biological pathway or process:

stemness (promotes); proliferation (promotes); other pathway/process (promotes)

Detected method:

Validation methods:

RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Transfection; Western Blot; Luciferase Reporter Assay; ChIP / ChIP-seq; In Vivo Animal Model; Bioinformatics Analysis

Clinical significance:

Description:

circRPPH1 is up-regulated in glioma cells, glioma cell spheres, and glioma tissues based on RT-qPCR and online dataset analysis. Functionally, circRPPH1 promotes glioma stemness, including CSC marker expression, sphere formation, ALDH activity, and tumor-initiating capacity in vivo. Mechanistically, c-FOS binds the circRPPH1 promoter and transcriptionally activates circRPPH1 expression, forming a c-FOS/circRPPH1 regulatory axis.

Confidence score:

0.7281
