circRNA-disease information
Expression pattern:
UP
Associated gene: RRM2
Associated microRNA: miR-593-5p
Biological function: Promotes proliferation, migration and aerobic glycolysis and inhibits apoptosis in gastric cancer cells under CoCl2-induced hypoxia; contributes to tumor growth in vivo.
Molecular mechanism: Acts as a ceRNA by binding miR-593-5p to regulate RRM2, affecting glycolysis-related proteins (e.g., HIF-1alpha, HK2, ENO1, LDHA, GLUT1, PKM2) and CoCl2-induced malignant phenotypes.
Biological pathway or process:

proliferation (promotes); migration (promotes); apoptosis (inhibits); cell cycle (other); glycolysis (promotes); ceRNA regulation (promotes)

Detected method:
Q
S
Validation methods:

Bioinformatics Analysis; RNA-seq; RT-qPCR; Clinical Sample Validation; Luciferase Reporter Assay; Transfection; CCK8; Wound Healing Assay; Transwell Assay; Annexin V/PI Flow Cytometry; Cell Cycle Assay; Western Blot; In Vivo Animal Model; IHC (Immunohistochemistry)

Clinical significance:

Potential independent prognostic biomarker and therapeutic target for gastric cancer.

Description:

circ_0043256 is up-regulated in gastric cancer tissues and GC cell lines. Its knockdown suppresses CoCl2-induced proliferation, migration/invasion, and aerobic glycolysis while promoting apoptosis and G2/M arrest. Mechanistically, circ_0043256 binds miR-593-5p to regulate the miR-593-5p/RRM2 axis and downstream glycolysis-related proteins, and its silencing inhibits tumor growth in xenograft models.

Confidence score:

0.7375
