circRNA-disease information
Expression pattern:
UP
Associated gene: Stat3, Snd1, Ptbp1
Associated microRNA: miRNA‐124, miR‐193a‐5p, miR‐192, miR‐561
Biological function: promotes macrophage pyroptosis; promotes inflammation and MMP synthesis; accelerates AAA formation and rupture; inhibits autophagy
Molecular mechanism: binds Stat3 to enhance NLRP3 transcription/levels; binds Snd1 and promotes Ptbp1 mRNA degradation leading to autophagy inhibition and reduced NLRP3 inflammasome degradation
Biological pathway or process:

pyroptosis (promotes); inflammation (promotes); autophagy (inhibits); ceRNA regulation (other); other pathway/process (other)

Detected method:

Validation methods:

RT-qPCR; ISH (In Situ Hybridization); RNase R Treatment; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Transfection; Western Blot; IF (Immunofluorescence); RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; Actinomycin D / DRB Stability Assay; In Vivo Animal Model; H&E Staining; ELISA; TUNEL; Flow Cytometry(Non-apoptosis/cycle); Bioinformatics Analysis

Clinical significance:

Description:

CircHipk3 is up-regulated in AAA aortic tissue and primarily localizes to the macrophage cytoplasm. It promotes macrophage pyroptosis and AAA progression by interacting with Stat3 to increase NLRP3 inflammasome production and by binding Snd1 to affect Ptbp1 mRNA degradation, inhibiting autophagy and reducing NLRP3 clearance.

Confidence score:

0.8355

Other information
Title:

CircHipk3 serves a dual role in macrophage pyroptosis by promoting NLRP3 transcription and inhibition of autophagy to induce abdominal aortic aneurysm formation.
