circRNA-disease information
Expression pattern:
UP
Associated gene: RNF44
Associated microRNA: hsa‐miR‐296‐5p
Biological function: Promotes proliferation, colony formation, migration, invasion, EMT and tumorigenicity; inhibits apoptosis; affects cell cycle progression in NSCLC cells.
Molecular mechanism: Acts as a ceRNA/miRNA sponge for miR-296-5p, relieving repression of RNF44.
Biological pathway or process:

ceRNA regulation (promotes); proliferation (promotes); migration (promotes); invasion (promotes); EMT (promotes); apoptosis (inhibits); cell cycle (other); metastasis (promotes)

Validation methods:

Microarray; Bioinformatics Analysis; RT-qPCR; RNase R Treatment; Nuclear-Cytoplasmic Fractionation; RNA Pull-Down; Luciferase Reporter Assay; Transfection; CCK8; Colony Formation Assay; Cell Cycle Assay; Annexin V/PI Flow Cytometry; Wound Healing Assay; Transwell Assay; Western Blot; In Vivo Animal Model; IHC (Immunohistochemistry); Clinical Sample Validation; Cohort Study; Survival Analysis

Clinical significance:

High expression of circ_515 was associated with worse survival of patients and correlated with advanced tumor node metastasis stage, poor tumor differentiation, and increased lymph node metastasis.

Description:

In NSCLC, circ_515 is up-regulated and promotes malignant phenotypes (proliferation, migration/invasion, EMT, tumor growth) while inhibiting apoptosis. Mechanistically, it mainly acts as a cytoplasmic ceRNA that sponges miR-296-5p to de-repress RNF44, forming a circ_515/miR-296-5p/RNF44 axis. High circ_515 expression is associated with worse patient survival and more aggressive clinicopathological features.

Confidence score:

0.8498
