| Associated gene: |
Pak2, Ago2, IRE1alpha, XBP1 |
| Associated microRNA: |
miR-130b-5p |
| Biological function: |
Promotes neurological recovery and neuronal survival after ischemic stroke rehabilitation; promotes protective ER stress; suppresses neuronal apoptosis. |
| Molecular mechanism: |
Acts as a miRNA sponge (ceRNA) for miR-130b-5p to disinhibit Pak2, enhancing IRE1alpha/XBP1 signaling and protective ER stress, thereby reducing apoptosis. |
| Biological pathway or process: |
apoptosis (inhibits); ceRNA regulation (promotes); other pathway/process (promotes) |

| Clinical significance: |
Plasma circAnks1b was higher in stroke patients receiving PE than sedentary patients and correlated with better neurological recovery; ROC analysis indicated high serum circAnks1b due to PE predicted better outcome (AUC=0.9352). |
| Description: |
In tMCAO ischemic stroke models, physical exercise upregulates circAnks1b in peri-infarct cortex. circAnks1b sponges miR-130b-5p to relieve repression of Pak2, enhancing IRE1alpha/XBP1-associated protective ER stress and suppressing neuronal apoptosis, improving neurological recovery. Plasma circAnks1b increases with exercise in IS patients and is associated with better functional outcome. |
