circRNA-disease information
Expression pattern:
UP
Associated gene: CIRBP, YAP1, EIF4A3, CRIM1 pre-mRNA, EIF4A4
Associated microRNA: miR-383-5p
Biological function: promotes proliferation; inhibits apoptosis; promotes tumor growth in vivo
Molecular mechanism: ceRNA sponge of miR-383-5p to upregulate CIRBP and activate PI3K/AKT signaling; hypoxia-enhanced YAP1-EIF4A3 interaction promotes circ_0007386 biogenesis by affecting CRIM1 pre-mRNA splicing/circularization
Biological pathway or process:

proliferation (promotes); apoptosis (inhibits); PI3K/AKT (promotes); ceRNA regulation (promotes)

Detected method:

Validation methods:

RNA-seq; RT-qPCR; Back-Splice Junction PCR / divergent primers PCR; Sanger Sequencing; RNase R Treatment; Actinomycin D / DRB Stability Assay; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Transfection; CCK8; Colony Formation Assay; TUNEL; Annexin V/PI Flow Cytometry; Luciferase Reporter Assay; Western Blot; RIP (RNA Immunoprecipitation); Co-IP; IHC (Immunohistochemistry); In Vivo Animal Model; H&E Staining; Survival Analysis; Bioinformatics Analysis

Clinical significance:

Higher expression of circ_0007386 is associated with worse overall survival and correlates with clinical features in NSCLC patients.

Description:

circ_0007386 is up-regulated in NSCLC and predominantly localized in the cytoplasm. It promotes NSCLC cell proliferation and tumor growth and inhibits apoptosis by sponging miR-383-5p to increase CIRBP, thereby activating PI3K/AKT signaling. Under hypoxia, enhanced YAP1-EIF4A3 interaction favors CRIM1 back-splicing and increases circ_0007386 biogenesis; high circ_0007386 predicts poorer overall survival.

Confidence score:

0.8933

Other information
Title:

Hypoxia-enhanced YAP1-EIF4A3 interaction drives circ_0007386 circularization by competing with CRIM1 pre-mRNA linear splicing and promotes non-small cell lung cancer progression.
