circRNA-disease information
Expression pattern:
UP
Associated gene: IMPDH2, hnRNPF, Alu elements, Alu Sc, Alu Jb
Associated microRNA: -
Biological function: promotes PCa cell proliferation in vitro and in vivo; promotes colony formation, migration and invasion; accelerates G1/S cell cycle transition
Molecular mechanism: circPFKP interacts with IMPDH2 and enhances the IMPDH2/guanine nucleotides axis; hnRNPF promotes circPFKP generation by binding flanking Alu elements
Biological pathway or process:

proliferation (promotes); cell cycle (promotes); migration (promotes); invasion (promotes); other pathway/process (promotes)

Detected method:
Q
M
Validation methods:

Microarray; RT-qPCR; Clinical Sample Validation; Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Actinomycin D / DRB Stability Assay; FISH / smFISH; Transfection; EdU Staining; Colony Formation Assay; Cell Cycle Assay; Transwell Assay; RNA Pull-Down; RIP (RNA Immunoprecipitation); Western Blot; In Vivo Animal Model; Bioinformatics Analysis

Clinical significance:

high circPFKP expression was correlated with higher D'Amico risk classification, higher N stage, and higher prognostic stage group, suggesting potential poorer prognosis and diagnostic/therapeutic application in PCa

Description:

circPFKP is a PFKP-derived circRNA up-regulated in prostate cancer tissues and associated with adverse clinicopathological features. It promotes PCa cell proliferation, migration, invasion, and G1/S cell-cycle progression, at least partly by interacting with IMPDH2 and enhancing the IMPDH2/guanine nucleotide axis. Its biogenesis is promoted by flanking Alu elements and regulated by hnRNPF.

Confidence score:

0.8415
