circRNA-disease information
Expression pattern:
UP
Associated gene: YTHDC1, KIF5B, KIF5B mRNA, PTBP1
Associated microRNA: miR-641
Biological function: Promotes malignant proliferation of HCC cells; downregulation inhibits cell proliferation, cell viability, colony formation, EdU-positive cells, and xenograft tumor growth.
Molecular mechanism: m6A-modified circCCAR1 is exported from the nucleus to the cytoplasm by YTHDC1; circCCAR1 competitively binds miR-641 to enhance KIF5B expression and binds PTBP1 to stabilize KIF5B mRNA.
Biological pathway or process:

proliferation (promotes); ceRNA regulation (promotes); m6A modification (other); mRNA stability (promotes)

Detected method:
Q
Validation methods:

RNase R Treatment; Actinomycin D / DRB Stability Assay; RT-qPCR; MeRIP / MeRIP-seq; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; FISH / smFISH; IF (Immunofluorescence); Nuclear-Cytoplasmic Fractionation; Transfection; CCK8; EdU Staining; Colony Formation Assay; In Vivo Animal Model; IHC (Immunohistochemistry); Western Blot; Bioinformatics Analysis

Clinical significance:

May provide a new target for the treatment of HCC.

Description:

circCCAR1 is upregulated in HCC cells and promotes malignant proliferation in vitro and in a HCCLM3 xenograft model. Mechanistically, m6A-modified circCCAR1 is exported to the cytoplasm by YTHDC1, acts as a ceRNA for miR-641 to increase KIF5B expression, and binds PTBP1 to stabilize KIF5B mRNA. The study suggests circCCAR1 may be a potential therapeutic target for HCC.

Confidence score:

0.8415
