circRNA-disease information
Expression pattern:
UP
Associated gene: IL-10, STAT3
Associated microRNA: miR-379-3 p
Biological function: promotes macrophage M2 polarization and promotes CRC metastatic nodule formation (metastasis) via exosomal transfer
Molecular mechanism: Exosomal circPOLQ enters macrophages and sponges miR-379-3p to de-repress IL-10, activating STAT3 signaling and driving M2 polarization.
Biological pathway or process:

JAK/STAT (promotes); metastasis (promotes); migration (promotes); invasion (promotes); EMT (promotes); immune regulation (promotes); ceRNA regulation (promotes)

Detected method:

S
Validation methods:

RNA-seq; RT-qPCR; Back-Splice Junction PCR / divergent primers PCR; Actinomycin D / DRB Stability Assay; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; ISH (In Situ Hybridization); Clinical Sample Validation; Survival Analysis; Luciferase Reporter Assay; RNA Pull-Down; Transfection; Western Blot; CCK8; Transwell Assay; Flow Cytometry(Non-apoptosis/cycle); In Vivo Animal Model; H&E Staining; IHC (Immunohistochemistry); Bioinformatics Analysis

Clinical significance:

Higher circPOLQ expression was associated with poorer prognosis in patients with CRC.

Description:

circPOLQ is up-regulated in colorectal cancer tissues and enriched in CRC cell-derived exosomes. After being transferred into macrophages, circPOLQ sponges miR-379-3p to increase IL-10 and activate STAT3 signaling, promoting M2 polarization and enhancing CRC invasion/metastatic nodule formation. High circPOLQ expression correlates with poorer overall survival in CRC patients.

Confidence score:

0.8693
