circRNA-disease information
Expression pattern:
UP
Associated gene: EIF4A3, ATG5, ATG7, USP14, Ago2
Associated microRNA: miR-3126-5p
Biological function: promotes CRC cell growth/proliferation and tumorigenesis; promotes autophagy; inhibits apoptosis
Molecular mechanism: ceRNA sponging of miR-3126-5p to upregulate EIF4A3; positive feedback where EIF4A3 promotes circEIF4A3 formation; EIF4A3 stabilizes ATG5 mRNA and stabilizes USP14 mRNA to enhance ATG7 protein stability
Biological pathway or process:

autophagy (promotes); proliferation (promotes); apoptosis (inhibits); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; divergent primers PCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; FISH / smFISH; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; Transfection; CCK8; EdU Staining; Colony Formation Assay; TUNEL; Annexin V/PI Flow Cytometry; Western Blot; In Vivo Animal Model; IHC (Immunohistochemistry)

Description:

circEIF4A3 (hsa_circ_0006749) is upregulated in colorectal cancer cells and promotes proliferation/tumor growth while reducing apoptosis. Mechanistically, it sponges miR-3126-5p to increase EIF4A3, forming a positive feedback loop in which EIF4A3 also enhances circEIF4A3 biogenesis; elevated EIF4A3 promotes autophagy by stabilizing ATG5 mRNA and USP14 mRNA to increase ATG7 protein stability.

Confidence score:

0.8378
