circRNA-disease information
Expression pattern:
DN
Associated gene: QKI
Associated microRNA: miR-224-5p, miR-1250-3p, miR-338-5p
Biological function: Inhibits cadmium-induced transformation of BEAS-2B cells; inhibits proliferation, EMT, invasion, migration, and anchorage-independent growth; promotes QKI expression.
Molecular mechanism: circ-SHPRH acts as a miR-224-5p sponge to regulate QKI expression, and QKI and circ-SHPRH form a positive-feedback loop under cadmium treatment.
Biological pathway or process:

proliferation (inhibits); EMT (inhibits); invasion (inhibits); migration (inhibits); ceRNA regulation (other); other pathway/process (inhibits)

Detected method:
Q
Validation methods:

RNase R Treatment; RT-qPCR; FISH / smFISH; RNA Pull-Down; Luciferase Reporter Assay; Transfection; Colony Formation Assay; Transwell Assay; Western Blot; Bioinformatics Analysis

Clinical significance:

Description:

circ-SHPRH is down-regulated during the middle and late stages of cadmium-induced transformation of human bronchial epithelial BEAS-2B cells. Functionally, circ-SHPRH suppresses proliferation, EMT, migration, invasion, and anchorage-independent growth by sponging miR-224-5p to restore QKI expression. The study also supports a positive-feedback loop between QKI and circ-SHPRH under cadmium treatment, suggesting circ-SHPRH as a potential target for preventing or treating cadmium-induced lung cancer.
