circRNA-disease information
Expression pattern:
UP
Associated gene: FAM83A, JNK, p38, ERK
Associated microRNA: miR-330-5p
Biological function: Circ-ZKSCAN1 promotes NSCLC progression by enhancing tumor cell proliferation, colony formation, migration, invasion and xenograft tumor growth, while inhibiting apoptosis; high expression is associated with larger tumor size, advanced clinical stage and poor prognosis.
Molecular mechanism: Circ-ZKSCAN1 functions as a ceRNA/miRNA sponge for miR-330-5p, thereby relieving miR-330-5p-mediated repression of FAM83A; FAM83A elevation feeds back to promote circ-ZKSCAN1 expression and the circ-ZKSCAN1/miR-330-5p/FAM83A loop inhibits MAPK/ERK signaling.
Biological pathway or process:

MAPK (inhibits); ERK (inhibits); proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); ceRNA regulation (promotes); other pathway/process (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Transfection; CCK8; Colony Formation Assay; Transwell Assay; Wound Healing Assay; Annexin V/PI Flow Cytometry; In Vivo Animal Model; IHC (Immunohistochemistry); RNA Pull-Down; Luciferase Reporter Assay; Western Blot; Survival Analysis; Bioinformatics Analysis

Clinical significance:

Circ-ZKSCAN1 is significantly up-regulated in NSCLC and is associated with malignant characteristics, poor prognosis, tumor size and clinical stage; it may serve as a potential biomarker and therapeutic target.

Description:

Circ-ZKSCAN1 (hsa_circ_0001727), derived from ZKSCAN1, is up-regulated in NSCLC tissues and cell lines and high expression is linked to larger tumors, advanced clinical stage and poorer survival. Functionally, it promotes NSCLC proliferation, migration, invasion and xenograft tumor growth and suppresses apoptosis. Mechanistically, circ-ZKSCAN1 sponges miR-330-5p to increase FAM83A expression, forming a feedback loop that inhibits MAPK/ERK signaling and promotes NSCLC progression.

Confidence score:

0.7794
