circRNA-disease information
Expression pattern:
UP
Associated gene: ALKBH5, YTHDF2
Associated microRNA: miR-486-3p, miR-409-3p
Biological function: associated with increased Th17% and increased autophagy level in CD4+T cells in RA
Molecular mechanism: hypomethylated (reduced m6A) circFOXK2; ALKBH5 regulates m6A level and circFOXK2 expression; predicted ceRNA binding to miR-486-3p and miR-409-3p
Biological pathway or process:

autophagy (promotes); immune regulation (other); ceRNA regulation (other)

Detected method:
Q
Validation methods:

Microarray; MeRIP / MeRIP-seq; RT-qPCR; Clinical Sample Validation; ROC Analysis; Bioinformatics Analysis; Transfection; Western Blot

Clinical significance:

circFOXK2 shows diagnostic value for distinguishing new-onset RA from HC and from revisiting RA (ROC AUC 0.631 and 0.706)

Description:

In peripheral blood CD4+ T cells from new-onset RA patients, circFOXK2 is up-regulated while its m6A-methylated level is decreased. circFOXK2 level correlates positively with Th17% and autophagy, and ALKBH5 modulates circFOXK2 m6A level and expression; bioinformatics predicts circFOXK2 may sponge miR-486-3p/miR-409-3p and has modest diagnostic value by ROC analysis.

Confidence score:

0.7039
