circRNA-disease information
Expression pattern:
UP
Associated gene: WNT1, beta-catenin, TCF4
Associated microRNA: -
Biological function: circ_001569 promotes NSCLC cell proliferation; knockdown of circ_001569 inhibits proliferation and colony formation; high circ_001569 expression is associated with poor survival and aggressive clinicopathological features.
Molecular mechanism: circ_001569 promotes NSCLC cell proliferation by regulating the Wnt/beta-catenin pathway; circ_001569 knockdown reduces mRNA and protein expression levels of WNT1, beta-catenin and TCF4.
Biological pathway or process:

Wnt/beta-catenin (promotes); proliferation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Transfection; CCK8; Colony Formation Assay; Western Blot; Survival Analysis

Clinical significance:

Increased circ_001569 expression is associated with tumor differentiation, lymph node metastasis and TNM classification, and high expression predicts poorer survival outcome in NSCLC.

Description:

circ_001569 is up-regulated in NSCLC tissues and cell lines and its high expression is associated with tumor differentiation, lymph node metastasis, TNM classification and poor survival. Functionally, circ_001569 promotes NSCLC cell proliferation, while siRNA-mediated knockdown suppresses proliferation and colony formation. Mechanistically, circ_001569 appears to activate the Wnt/beta-catenin pathway by maintaining WNT1, beta-catenin and TCF4 mRNA/protein expression.

Confidence score:

0.6148
