Associated microRNA: miR-22, miR-136*, miR-138, miR-149*, miR-433, miR-762, miR-3614-5p, miR-3622b-5p
Biological function: Suppresses tumor growth, cancer cell proliferation and survival; decreases cell viability; promotes apoptosis/cell death; inhibits blood vessel formation/angiogenesis and local invasion; increases Foxo3 expression.
Molecular mechanism: circ-Foxo3 functions as a miRNA sponge for eight miRNAs, competitively reducing miRNA binding to Foxo3 mRNA and thereby promoting Foxo3 translation and protein expression.

Description:

In this study, circ-Foxo3 is a Foxo3-derived circular RNA that is lower in breast tumor/cancer contexts than in normal tissues or non-cancer cell lines. Ectopic circ-Foxo3 suppresses breast cancer cell proliferation/survival and xenograft tumor growth, promotes apoptosis, reduces CD34-associated vascularization, and inhibits local invasion. Mechanistically, circ-Foxo3 acts as a sponge for eight miRNAs, relieving repression of Foxo3 mRNA translation and increasing Foxo3 protein expression.
