circRNA-disease information
Expression pattern:
UP
Associated gene: RACK1
Associated microRNA: -
Biological function: promotes fibroblast proliferation, migration, and ECM deposition; inhibits apoptosis; exacerbates inflammatory response and oxidative stress in keloid
Molecular mechanism: circFoxp1 is cytoplasmic and interacts with the RNA-binding protein RACK1; circFoxp1 overexpression is associated with reduced RACK1 levels and enhanced inflammation/oxidative stress-related programs.
Biological pathway or process:

proliferation (promotes); migration (promotes); apoptosis (inhibits); inflammation (promotes); oxidative phosphorylation (other); cell cycle (promotes); other pathway/process (promotes)

Detected method:

S
Validation methods:

RT-qPCR; RNA-seq; FISH / smFISH; ISH (In Situ Hybridization); Sanger Sequencing; Transfection; CCK8; Colony Formation Assay; Transwell Assay; Annexin V/PI Flow Cytometry; RNA Pull-Down; Western Blot; In Vivo Animal Model; IHC (Immunohistochemistry); H&E Staining

Clinical significance:

circFoxp1 may serve as a novel, promising therapeutic target for keloid

Description:

circFoxp1 is up-regulated in human keloid tissues and mainly localizes to the cytoplasm. Its overexpression promotes fibroblast proliferation, migration/invasion and ECM deposition while inhibiting apoptosis, and it can aggravate keloid growth in a patient-derived xenograft nude-mouse implantation model. Mechanistically, circFoxp1 interacts with the RNA-binding protein RACK1 and is associated with inflammatory/oxidative-stress related changes.
