circWNK1 (hsa_circ_0003251), derived from WNK1, is down-regulated in gastric cancer tissues/cell lines and acts as a tumor suppressor. It sponges miR-21-3p to restore SMAD7 expression, thereby inhibiting TGF-beta/SMAD signaling, leading to reduced proliferation, migration, invasion, EMT, tumor growth and metastasis. Low circWNK1 expression is linked to worse overall survival and has diagnostic potential (AUC 0.714).
