| Biological function: |
Inhibits gastric cancer cell proliferation and suppresses tumor growth. |
| Molecular mechanism: |
circIPO7 directly binds caprin-1, competes with G3BP1, prevents recruitment of caprin-1 and its target mRNAs (EGFR and mTOR) to ribosomes, thereby inhibiting their translation and inactivating PI3K/AKT/mTOR signaling. |

RNA-seq; RT-qPCR; divergent primers PCR; RNase R Treatment; Sanger Sequencing; FISH / smFISH; Clinical Sample Validation; In Vivo Animal Model; Transfection; CCK8; EdU Staining; Colony Formation Assay; RNA Pull-Down; RIP (RNA Immunoprecipitation); Co-IP; Western Blot; Bioinformatics Analysis |
| Clinical significance: |
Independent prognostic factor; lower circIPO7 expression may have shorter recurrence-free survival (RFS) and overall survival (OS). |
| Description: |
circIPO7 is down-regulated in gastric cancer and functions as a tumor suppressor by inhibiting cell proliferation and xenograft growth. Mechanistically, circIPO7 binds the RBP caprin-1, competitively disrupts caprin-1-G3BP1 interaction, prevents caprin-1/target mRNAs (EGFR, mTOR) from associating with ribosomes, and suppresses PI3K/AKT/mTOR signaling via translational inhibition of EGFR and mTOR. |
