circRNA-disease information
Expression pattern:
DN
Associated gene: GLIS2 mRNA, EIF4A3, DNMT1
Associated microRNA: -
Biological function: Suppresses HCC cell proliferation, migration, invasion, EMT, tumour growth and metastasis, and promotes cancer cell apoptosis.
Molecular mechanism: GLIS2 promoter hypermethylation reduces circ_GLIS2 expression in HCC; EIF4A3 directly binds GLIS2 mRNA transcript and promotes circ_GLIS2 and GLIS2 expression; circ_GLIS2 overexpression suppresses EMT-related marker changes.
Biological pathway or process:

proliferation (inhibits); migration (inhibits); invasion (inhibits); apoptosis (promotes); EMT (inhibits); metastasis (inhibits); mRNA stability (other); other pathway/process (other)

Detected method:
Q
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Actinomycin D / DRB Stability Assay; RT-qPCR; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RIP (RNA Immunoprecipitation); RNA Pull-Down; Transfection; CCK8; Colony Formation Assay; Annexin V/PI Flow Cytometry; Wound Healing Assay; Transwell Assay; In Vivo Animal Model; H&E Staining; IHC (Immunohistochemistry); IF (Immunofluorescence); Western Blot; Survival Analysis; ROC Analysis; Bioinformatics Analysis; MSP (Methylation-Specific PCR)

Clinical significance:

Low circ_GLIS2 levels correlated with poor survival; circ_GLIS2 level discriminated advanced-stage from early-stage HCC patients and could serve as a diagnosis marker or therapeutic target for HCC.

Description:

circ_GLIS2 is a human GLIS2-derived circRNA that is down-regulated in HCC tissues and cell lines. Its overexpression suppresses HCC proliferation, migration, invasion, EMT, tumor growth and metastasis while promoting apoptosis, and its low expression is associated with poorer survival. Mechanistically, reduced circ_GLIS2 in HCC is linked to GLIS2 promoter hypermethylation, while EIF4A3 positively regulates circ_GLIS2/GLIS2 expression through binding the GLIS2 transcript.
