circRNA-disease information
Expression pattern:
UP
Associated gene: STAT1
Associated microRNA: miR-1296
Biological function: promotes M1 macrophage polarization, increases the M1/M2 ratio, alleviates cisplatin resistance in gastric cancer cells, reduces GC cell viability under cisplatin treatment, and increases cisplatin-induced apoptosis
Molecular mechanism: circSOD2 acts as a microRNA sponge for miR-1296 and regulates STAT1 expression through the miR-1296/STAT1 axis, thereby promoting M1 macrophage polarization and enhancing cisplatin sensitivity
Biological pathway or process:

ceRNA regulation (promotes); macrophage polarization (promotes); immune regulation (promotes); apoptosis (promotes); drug resistance (inhibits); chemoresistance (inhibits)

Detected method:
Q
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; RT-qPCR; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RNA Pull-Down; Luciferase Reporter Assay; Transfection; CCK8; Annexin V/PI Flow Cytometry; Flow Cytometry(Non-apoptosis/cycle); ELISA; Western Blot; In Vivo Animal Model; Bioinformatics Analysis

Clinical significance:

CircSOD2 may serve as a potential target for GC chemotherapy and may be an applicable diagnostic or prognostic biomarker for immune therapy

Description:

CircSOD2 is up-regulated in M1 macrophages and promotes M1 macrophage polarization in gastric cancer-related macrophage models. Mechanistically, cytoplasmic circSOD2 sponges miR-1296 to regulate STAT1, increasing the M1/M2 ratio and enhancing cisplatin-induced apoptosis while reducing cisplatin resistance in gastric cancer cells.

Confidence score:

0.8278
