circRNA-disease information
Expression pattern:
UP
Associated gene: AZGP1, c-Met, G6PD, mTOR, SREBP1, YY1, HuR, AGO2, NOLC1
Associated microRNA: miR-212-5p
Biological function: promotes colorectal cancer cell proliferation, migration, and liver metastasis; promotes oxPPP reprogramming and increases G6PD activity; supports NADPH production and reduces ROS
Molecular mechanism: Interacts with AZGP1 to activate mTOR/SREBP1 signaling; sponges miR-212-5p to upregulate c-Met, inducing G6PD and activating oxPPP; regulated by YY1; stabilizes host gene NOLC1 mRNA via HuR
Biological pathway or process:

PI3K/AKT/mTOR (promotes); oxidative phosphorylation (other); mitochondrial function (other); proliferation (promotes); migration (promotes); metastasis (promotes); ceRNA regulation (promotes); other pathway/process (promotes)

Clinical significance:

circNOLC1 signal was increased in liver metastasis compared with corresponding primary CRC and was correlated with higher SREBP1, G6PD and c-Met and lower miR-212-5p in human specimens

Description:

circNOLC1 is up-regulated in CRC, especially in liver metastasis, and promotes CRC cell proliferation, migration, and liver metastatic colonization. Mechanistically, circNOLC1 binds AZGP1 to activate mTOR/SREBP1 signaling and also sponges miR-212-5p to increase c-Met, converging on G6PD activation and oxidative PPP reprogramming with increased NADPH/ROS buffering.

Confidence score:

0.8665
