circRNA-disease information
Expression pattern:
DN
Associated gene: S1PR1, AGO2, TNF-alpha, IL-6, STAT3
Associated microRNA: miR-4689
Biological function: promotes proliferation; inhibits apoptosis; modulates T-cell activation; inhibits secretion of TNF-alpha and IL-6
Molecular mechanism: ceRNA mechanism (miRNA sponge) targeting miR-4689 to regulate S1PR1 expression
Biological pathway or process:

ceRNA regulation (promotes); proliferation (promotes); apoptosis (inhibits); JAK/STAT (other); inflammation (inhibits)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; FISH / smFISH; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Transfection; CCK8; Annexin V/PI Flow Cytometry; Cell Cycle Assay; Western Blot; ELISA; Bioinformatics Analysis

Clinical significance:

Lower circPTPN22 expression was associated with higher SLEDAI scores, and circPTPN22 may be a potential biomarker for SLE.

Description:

circPTPN22 is down-regulated in PBMCs from SLE patients and is associated with disease activity. In Jurkat T cells, circPTPN22 promotes proliferation and inhibits apoptosis, and suppresses TNF-alpha/IL-6 secretion by acting as a miR-4689 sponge to de-repress S1PR1 (circPTPN22/miR-4689/S1PR1 axis), with links to JAK-STAT-related signaling.

Confidence score:

0.7295
