circRNA-disease information
Expression pattern:
UP
Associated gene: PGF, YTHDF1, METTL3
Associated microRNA: miR-342-5p
Biological function: promotes colorectal cancer cell migration and invasion and promotes metastasis
Molecular mechanism: m6A modification enhances circALG1 binding to miR-342-5p and promotes ceRNA function to upregulate PGF; YTHDF1-mediated m6A recognition contributes by enhancing circALG1 stability
Biological pathway or process:

metastasis (promotes); migration (promotes); invasion (promotes); EMT (promotes); MAPK (promotes); ERK (promotes); ceRNA regulation (promotes); m6A modification (promotes)

Detected method:
Q
M
Validation methods:

Microarray; RT-qPCR; Back-Splice Junction PCR / divergent primers PCR; Sanger Sequencing; RNase R Treatment; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; FISH / smFISH; Transfection; Transwell Assay; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; MeRIP / MeRIP-seq; Western Blot; RNA-seq; Bioinformatics Analysis; In Vivo Animal Model; H&E Staining; Clinical Sample Validation; ROC Analysis

Clinical significance:

circALG1 expression distinguished CRC tumours vs adjacent tissues and CRC peripheral blood vs healthy individuals (ROC analysis); high circALG1 expression associated with metastasis and higher CA19-9; proposed prognostic marker/therapeutic target

Description:

circALG1 is up-regulated in colorectal cancer tissues and peripheral blood and promotes CRC metastasis by enhancing migration and invasion. Mechanistically, m6A-modified circALG1 (recognized by YTHDF1) sponges miR-342-5p to upregulate PGF, which activates ERK/MAPK and EMT-associated changes.

Confidence score:

0.8861
