circRNA-disease information
Expression pattern:
UP
Associated gene: OXSR1
Associated microRNA: miR-339-5p
Biological function: CircNRIP1 promotes LPS-induced HK2 cell apoptosis and inflammation; circNRIP1 knockdown alleviates LPS-induced HK2 cell injury by improving cell viability and reducing apoptosis and inflammatory cytokine production.
Molecular mechanism: CircNRIP1 acts through a ceRNA-like circNRIP1/miR-339-5p/OXSR1 pathway, binding miR-339-5p and positively regulating OXSR1 expression.
Biological pathway or process:

apoptosis (promotes); inflammation (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RNase R Treatment; RT-qPCR; Clinical Sample Validation; RNA Pull-Down; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Bioinformatics Analysis; Nuclear-Cytoplasmic Fractionation; Transfection; CCK8; Annexin V/PI Flow Cytometry; Western Blot; ELISA

Clinical significance:

CircNRIP1 was upregulated in blood samples from septic AKI patients and may be a therapeutic target for AKI incurred by sepsis.

Description:

CircNRIP1 is up-regulated in blood from septic AKI patients and in LPS-induced HK2 cells. The study shows that circNRIP1 knockdown protects HK2 cells from LPS-induced injury by reducing apoptosis and inflammatory cytokine production, mechanistically through the miR-339-5p/OXSR1 axis.

Confidence score:

0.7968
