circRNA-disease information
Expression pattern:
DN
Associated gene: IGF2BP1, CCL5 mRNA
Associated microRNA: -
Biological function: CircMAPK1 suppresses LUAD tumor growth and promotes intratumoral CD8 + T cell infiltration/recruitment.
Molecular mechanism: CircMAPK1 binds IGF2BP1 in the cytoplasm and induces IGF2BP1 occupancy on the 3′UTR of CCL5 mRNA, thereby maintaining CCL5 mRNA stability and promoting CCL5-mediated CD8 + T cell recruitment.
Biological pathway or process:

immune regulation (promotes); mRNA stability (promotes); proliferation (inhibits)

Detected method:

S
Validation methods:

RNA-seq; RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; Transfection; Transwell Assay; Flow Cytometry(Non-apoptosis/cycle); In Vivo Animal Model; IHC (Immunohistochemistry); ELISA; Western Blot; Bioinformatics Analysis

Clinical significance:

CircMAPK1 is positively associated with CD8 + T cell infiltration and is proposed as a potential nucleic acid drug to improve immunotherapy response efficiency combined with ICB.

Description:

CircMAPK1 is down-regulated in LUAD and acts as an anti-tumor microenvironment-associated circRNA. It binds IGF2BP1 in the cytoplasm, promotes IGF2BP1-dependent stabilization of CCL5 mRNA, increases CCL5 expression, and thereby enhances CD8 + T cell recruitment/infiltration. The study suggests circMAPK1 may be a nucleic acid drug candidate to improve ICB immunotherapy response.

Confidence score:

0.8658
