circRNA-disease information
Expression pattern:
UP
Associated gene: AR, SLCO1B7, NcoR1, TPD52, RISC
Associated microRNA: miR-1295, miR-139-5p, miR-671-5p
Biological function: enhances invasion and decreases cisplatin response in lung cancer cells; promotes lung cancer progression
Molecular mechanism: AR transcriptionally represses circ-SLCO1B7 via binding to SLCO1B7 promoter ARE1/2 (with NcoR1); circ-SLCO1B7 acts as a miRNA sponge for miR-139-5p, relieving repression of TPD52
Biological pathway or process:

invasion (promotes); migration (promotes); drug resistance (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; RT-qPCR; RNA Pull-Down; Luciferase Reporter Assay; Transfection; MTT; Transwell Assay; Western Blot; ChIP / ChIP-seq; Co-IP; Clinical Sample Validation; Bioinformatics Analysis; In Vivo Animal Model; IHC (Immunohistochemistry); H&E Staining

Clinical significance:

circ-SLCO1B7 expression correlates with tumor size, T grade, N grade, and differentiation; increased in lung cancer tissues vs adjacent normal tissues

Description:

circ-SLCO1B7 is up-regulated in lung cancer tissues and promotes lung cancer cell invasion while reducing cisplatin response. Mechanistically, it acts as a ceRNA that sponges miR-139-5p to increase TPD52, and its transcription is repressed by AR via binding the SLCO1B7 promoter (ARE1/2, with NcoR1), forming an AR/circ-SLCO1B7/miR-139-5p/TPD52 axis validated in cells, clinical specimens, and an in vivo xenograft model.

Confidence score:

0.8494
