circRNA-disease information
Expression pattern:
UP
Associated gene: RAB3D, Ago2
Associated microRNA: miR-588
Biological function: promotes OS cell viability, proliferation, migration and invasion; inhibits apoptosis; promotes tumor growth in vivo
Molecular mechanism: circECE1 functions as a ceRNA by sponging miR-588 to regulate RAB3D expression
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); metastasis (promotes); ceRNA regulation (other)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Clinical Sample Validation; Transfection; CCK8; EdU Staining; Annexin V/PI Flow Cytometry; Transwell Assay; Wound Healing Assay; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); Western Blot; In Vivo Animal Model; IHC (Immunohistochemistry)

Clinical significance:

High expression of circECE1 was closely associated with advanced TNM stage, positive lymph node metastasis, and large tumor size

Description:

circECE1 (hsa_circ_0002402), derived from ECE1, is up-regulated in osteosarcoma tissues and cells and promotes malignant phenotypes (proliferation, migration/invasion) while inhibiting apoptosis. Mechanistically, circECE1 sponges miR-588 to de-repress RAB3D, forming a circECE1/miR-588/RAB3D ceRNA axis; circECE1 knockdown suppresses xenograft tumor growth.

Confidence score:

0.8172
