circRNA-disease information
Expression pattern:
UN
Associated gene: SP2, NAP1L1, AGO2
Associated microRNA: miR-770-5p
Biological function: promotes cisplatin chemoresistance and decreases susceptibility to NK cell-mediated cytotoxicity in OSCC cells
Molecular mechanism: SP2 transcriptionally induces circPUM1; circPUM1 acts as a miRNA sponge for miR-770-5p to regulate NAP1L1, thereby modulating cisplatin resistance and NK cell cytotoxicity in OSCC cells
Biological pathway or process:

immune regulation (other); chemoresistance (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; RT-qPCR; Nuclear-Cytoplasmic Fractionation; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Transfection; CCK8

Clinical significance:

-

Description:

In OSCC cell lines, circPUM1 promotes cisplatin (CDDP) chemoresistance and reduces susceptibility to NK cell-mediated killing. Mechanistically, SP2 transcriptionally upregulates circPUM1, which sponges miR-770-5p to de-repress NAP1L1, forming an SP2/circPUM1/miR-770-5p/NAP1L1 axis.

Confidence score:

0.7366
