circRNA-disease information
Expression pattern:
UP
Associated gene: SLC7A11, Ago2, GPX4
Associated microRNA: miR-1287-5p
Biological function: circCOL5A1 functions as an oncogenic circRNA in CRC; it promotes CRC cell proliferation, invasion, and tumor growth, and inhibits ferroptosis.
Molecular mechanism: circCOL5A1 acts as a ceRNA by binding miR-1287-5p, thereby regulating SLC7A11 expression and affecting CRC malignant phenotype and ferroptosis.
Biological pathway or process:

proliferation (promotes); invasion (promotes); ferroptosis (inhibits); ceRNA regulation (other)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Cohort Study; Survival Analysis; Bioinformatics Analysis; Transfection; CCK8; EdU Staining; Transwell Assay; IF (Immunofluorescence); Western Blot; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); RNA Pull-Down; In Vivo Animal Model; H&E Staining; IHC (Immunohistochemistry)

Clinical significance:

Increased circCOL5A1 levels were related to TNM stage, lymph node metastasis, distant metastasis, tumor differentiation, and low overall survival rate in CRC patients; circCOL5A1 might be a CRC biomarker.

Description:

circCOL5A1 is up-regulated in colorectal cancer tissues and cells and is associated with advanced clinicopathological features and poor overall survival. Functionally, circCOL5A1 promotes CRC cell proliferation, invasion, and xenograft tumor growth while suppressing ferroptosis. Mechanistically, it acts through the circCOL5A1/miR-1287-5p/SLC7A11 axis.

Confidence score:

0.8067
