circRNA-disease information
Expression pattern:
UP
Associated gene: CDKN2A mRNA, SMAD3 mRNA, YTHDC1, YTHDF2, AGO2
Associated microRNA: miR-1197
Biological function: promotes tumor cell proliferation and growth; promotes cell cycle progression; promotes tumor progression
Molecular mechanism: m6A-dependent cytoplasmic export via YTHDC1; direct circRNA-mRNA interaction promotes CDKN2A mRNA decay by recruiting YTHDF2; ceRNA sponging of miR-1197 to derepress SMAD3
Biological pathway or process:

proliferation (promotes); cell cycle (promotes); ceRNA regulation (promotes); m6A modification (promotes); mRNA stability (inhibits)

Detected method:
Q
H
Validation methods:

FISH / smFISH; RT-qPCR; Nuclear-Cytoplasmic Fractionation; Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); RNA Pull-Down; Transfection; CCK8; EdU Staining; Colony Formation Assay; Cell Cycle Assay; Flow Cytometry(Non-apoptosis/cycle); In Vivo Animal Model; H&E Staining; Western Blot; Bioinformatics Analysis; MeRIP / MeRIP-seq

Clinical significance:

high circMET correlates with higher tumor stage; link with prognosis not firmly established due to small sample size

Description:

In NONO-TFE3 tRCC, circMET is up-regulated and promotes tumor cell proliferation and cell-cycle progression. Mechanistically, m6A-modified circMET is exported to the cytoplasm via YTHDC1, directly binds CDKN2A mRNA to recruit YTHDF2 and accelerate CDKN2A mRNA decay, and acts as a ceRNA sponging miR-1197 to upregulate SMAD3.

Confidence score:

0.8165
