circRNA-disease information
Expression pattern:
UP
Associated gene: TXNIP, NLRP3, Caspase 1, ASC
Associated microRNA: miR-136-5p
Biological function: promotes LPS-induced HK2 cell death, oxidative stress, inflammatory release and NLRP3 inflammasome activation; circ_0001818 downregulation alleviates LPS-induced HK2 cell injury
Molecular mechanism: circ_0001818 functions as a molecular sponge of miR-136-5p to regulate TXNIP expression and activate the TXNIP/NLRP3 inflammasome pathway
Biological pathway or process:

ceRNA regulation (promotes); apoptosis (promotes); oxidative phosphorylation (other); inflammation (promotes); pyroptosis (promotes); other pathway/process (promotes)

Detected method:
Q
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; RT-qPCR; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RNA Pull-Down; Luciferase Reporter Assay; Transfection; CCK8; Annexin V/PI Flow Cytometry; ELISA; Western Blot; ROC Analysis; Bioinformatics Analysis

Clinical significance:

exosomal circ_0001818 has potential diagnostic value in septic AKI; AUC of circ_0001818 in sepsis exosomes was 0.792

Description:

circ_0001818 is up-regulated in LPS-treated HK2 septic AKI cell models and in serum-derived exosomes from sepsis/septic AKI patients. Functionally, circ_0001818 promotes LPS-induced renal tubular epithelial cell injury, including cell death, oxidative stress, inflammatory cytokine secretion and NLRP3 inflammasome activation, by sponging miR-136-5p and increasing TXNIP expression. Exosomal circ_0001818 also shows diagnostic biomarker potential for septic AKI.

Confidence score:

0.8562
