circRNA-disease information
Expression pattern:
UP
Associated gene: LARP1, Ago2
Associated microRNA: miR-330-5p
Biological function: Promotes NSCLC cell growth, migration/invasion and glutamine metabolism; inhibits apoptosis; promotes tumor growth in vivo.
Molecular mechanism: ceRNA mechanism: circ-PDZD8 sponges miR-330-5p to increase LARP1 expression.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); glutaminolysis (promotes); other pathway/process (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Nuclear-Cytoplasmic Fractionation; RNase R Treatment; Transfection; CCK8; Colony Formation Assay; Annexin V/PI Flow Cytometry; Transwell Assay; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); Western Blot; In Vivo Animal Model; H&E Staining; Clinical Sample Validation

Clinical significance:

Circ-PDZD8 high expression is associated with advanced TNM stage and lymph node metastasis (Table 1).

Description:

Circ-PDZD8 is up-regulated in NSCLC tissues and cell lines and mainly localizes to the cytoplasm. It promotes malignant phenotypes and glutamine metabolism by sponging miR-330-5p to increase LARP1 expression, thereby enhancing tumor growth in vitro and in a xenograft model.

Confidence score:

0.7637
